I have a box intake manifold that has the map sensor opening welded shut so I can just use the MAP sensor on Megasquirt.
My problem is that I need to pass inspection. If I got a vacuum block and modded a MAP sensor onto that, would it work and read correctly?

Yes it will work, as long as the vacuum hose isn't too long, or really small in diameter.
Otherwise there may be a risk of a MAP/TPS correlation code.
If you have a spare nipple off the plenum, you could just use a 1" long piece of vacuum tubing to connect it right to the plenum.
